Describe the change in the pathway during the initial absorption of light. Rh -> Rh* -> activates G protein transducin -> activates PDE -> conversion of cGMP to GMP -> closing of cGM P gated cation channel.
How does Ca2+ keep cGMP in check? 1). By activating calmodulin which directly inhibits the openning of the channel. 2). By activating GCAP which inhibits guanylate cyclase. Since Guanylate cyclase converts GTP to cGMP, this reduces the amount of cGMP. 3). By activating Recoverin, which inhibits Rh Kinase and Arrestin.
What are the methods of studying rods and cones? 1). Intracellular recording (50 years ago), 2). Whole-cell recording from intact cell or isolated outer segment (allows internal dialysis) and 3). Suction-pipette recording (stability, monitors selectively the memrane current associated with phototransduction in the outer segment)
