Name and describe 3 molecules involved in post-synaptic differentiation in central synapses. NARP induces clustering of AMPA receptors. Eph receptor tyrosine kinases potentially induces clustering of NMDA receptors. Cadherins, present on pre and postsynaptic membranes, may establish cytoskeletal stabilizations and specificity on pre and postsynaptic contact.
Describe three events that require interactions between the muscle and nerve that are required for the final NMJ. 1). Clustering of AChR at postsynaptic sites, which require agrin, Musk, and Rapsyn. 2). Dispersion of extrasynaptic AChR, which is dependent on electrical activity, ACh, and Cdk5. 3). What are the 3 sequential processes for the development of synapses? 1). Target recognition 2). synaptic differentiation and 3). stabilization of destabilization.
What are the three main events that characterizes the differentiation of the neuromuscular junction (classical view)? 1. Clustering of diffuse Ach Receptors at the postsynaptic membrane 2. Enhancement of AchR transcription at intrasynaptic sites and 3. Repression of AchR transcription at extrasynaptic sites.
Agrin GOF Injection of recombinant agrin into muscles or denervated muscle fibers results in patches of agrin on the surfice of muscle with postsynaptic apparatus.
Agrin LOF Agrin KO mice have postsynaptic differentiation impaired. Few AchR clusters.
What is the Agrin hypothesis? That agrin is the master nerve-derived initiator of postsynaptic differentiation.
What is the current hypothesis for the clustering of AchR?

First, Musk and rapsyn leads to the formation of protosynapses. The second step is nerve-mediated and requires neuronal agrin which mediates the refinement of the synapses by elimination of protosynapses.

Lrp4 A receptor for Agrin and forms a complex with MuSK.
What is the current speculation on the transcription of postsynaptic transcription of AChR? Agrin is responsible for transcription of AchRs in subsynaptic nuclei.
What molecule is important for the dispersal of AChR? Cdk5
What are some of the possible signals for synapse-specific elimination and/or strengthening? synaptotrophins, synaptotoxins or synaptomedins
